RealStar^® PCR Kit optimization for Clostridium difficile and West Nile Virus

RealStar^® Clostridium difficile PCR Kit 2.0 (Ref: 172013) for the detection and differentiation of Clostridium difficile toxin A and toxin B specific DNA.

The RealStar^® Clostridium difficile PCR Kit 2.0 was optimized in comparison to RealStar^® Clostridium difficile PCR Kit 1.0 to increase the performance of the kit and harmonized within the RealStar^® product line.

Please note: main change affects the temperature profile and the Internal Control amplification system.

Temperature profile Kit version 1.0: 120sec @95°C, for 45 cycles 15sec @95°C, 45sec @58°C

Temperature profile Kit version 2.0: 120sec @95°C, for 45 cycles 15sec @95°C, 45sec @58°C, 15sec @72°C

The following product is available as RUO:

RealStar^® WNV RT-PCR Kit 2.0 (Ref: 322003) for the detection of West Nile virus specific RNA.

The RealStar^® WNV RT-PCR Kit 2.0 was optimized in comparison to RealStar^® WNV RT-PCR Kit 1.0 to increase the performance of the kit especially in respect of the Internal Control amplification system.

Please note: main changes affect the pipetting scheme and the temperature profile.

• Reaction volume Kit version 1.0 => 50µL
• Reaction volume Kit version 2.0  => 30µL

• Temperature profile Kit version 1.0: 600sec @50°C, 120sec @95°C, for 45 cycles 15sec @95°C, 45sec @55°C, 15sec @72°C.
• Temperature profile Kit version 2.0: 1200sec @55°C, 120sec @95°C, for 45 cycles 15sec @95°C, 45sec @55°C, 15sec @72°C.

Additionally, for harmonization of the kits from the tropical panel the reference dye ROX was included to the RealStar^® WNV RT-PCR Kit 2.0. The CE mark is planned for 2019.
